In de game zul je met vader en zoon Rick en Alex O'Connell een gedaanteverwisselende mummie moeten stoppen, die herrezen is door een eeuwenoude vloek. Daarbij kom je op levensgevaarlijke locaties waaronder Chinese tombes en het Himalaya-gebergte.In de game zul je vechten met je vuisten en met vuurwapens tegen vijanden uit de film. Zo zul je onder andere een driekoppige Gorgon en terracotta krijgers tegenkomen. De gevechten worden afgewisseld door puzzels. Om geheime ruimtes en verborgen schatten te vinden zul je bijvoorbeeld hiërogliefen moeten ontcijferen.
